Abbkine線粒體膜電位測定試劑盒（Mitochondrial Membrane Potential Assay Kit , JC-1貨號KTA4001）提供了一種簡單方法，基於碳菁染料(carbocyanine dye ) JC-1通過檢測線粒體跨膜電位的變化來區分健康細胞和凋亡細胞(apoptotic cells)。在健康細胞中，這種染料在粒腺體中積累(accumulates)和聚集(aggregates)，形成鮮紅色的螢光團塊。耗散線粒體膜電位都會阻止JC-1染料在線粒體中積聚，因此該染料以其單體形式保留在細胞質中，導致從紅色的轉變（聚結的JC-1，Ex / Em = 585/590 nm）發出綠色螢光（JC-1單體，Ex / Em = 510/527 nm）。 JC-1可用作多種細胞類型（包括肌細胞和神經元）以及完整組織和分離的粒腺體中粒腺體潛力的指標。該試劑盒包含CCCP，CCCP導致質子梯度解偶聯，該質子梯度在電子傳輸鏈中電子載體的正常活動過程中建立，並因此消除了線粒體的電化學勢，可以用作防止JC-1聚集的對照組。

BrdU(5-Bromo-2-deoxyUridine) 或 5-bromodeoxyuridine 是一種胸苷核苷 (thymidine nucleoside , T) 類似物，可以競爭性取代胸苷核苷 T (thymidine nucleoside T)以摻入 DNA 合成階段（DNA synthesis phase , S 階段）。 Anti-brdu assay kits 將活躍分裂的細胞與(G4102)中的BrdU Anti-BrdU (mouse mAb)抗體混合，結合相應的抗體連接酶或螢光素作為指示系統，可檢測含有BrdU的細胞，結合與其他細胞標記物進行雙重標記，可以確定增殖細胞的類型、增殖率(proliferation rate)等，這對細胞動力學的研究具有重要意義。通過體內註射或細胞培養將 BrdU 引入組織細胞。 BRDU定位準確，能有效反映S期細胞新合成DNA(synthesized DNA)的水平。而且受細胞內外環境影響小，標記不會丟失。

Assay Protocol / Procedures

(動物實驗) 用生理鹽水溶解粉狀BrdU，配成10mg/mL濃度，過濾滅菌備用。一般以50mg/kg的劑量腹腔注射4次，每次間隔2小時。最後一次注射後 24 小時犧牲動物。

(細胞測定) 對於細胞，BrdU 的濃度和處理時間取決於細胞類型。一般來說，治療腫瘤細胞(tumor cells)所需的濃度和時間較低，對成纖維細胞(fibroblasts)或上皮細胞(epithelial cells)等增殖較慢的細胞則需提高濃度，延長作用時間。推薦濃度範圍為20-100 μM，作用時間為40 min-4 h，可根據具體細胞類型進行調整。下表僅供參考，列出了測試的普通細胞處理的濃度和時間。

Wst-8 was an upgrade of MTT and was reduced by mitochondrial dehydrogenase in the presence of electron-coupled reagents to form a highly water-soluble orange dirty product (Formazan). Wst-8是MTT的升級版，在電子偶聯試劑存在下被線粒體脫氫酶還原，形成高度水溶性的產物（Formazan）。CCK-8 通過使用高度水溶性四唑鹽-WST-8 允許非常方便的測定。 WST-8 在細胞中被脫氫酶(dehydrogenases)還原，得到一種可溶於組織培養基的橙色產物（formazan）。 CCK-8 是非放射性的，允許進行靈敏的比色測定，以確定細胞增殖(cell proliferation)和細胞毒性(cytotoxicity assays)測定中的活細胞數量。

CheKine™活性氧（CheKine™ Reactive Oxygen Species , ROS）檢測分析試劑盒提供了一種簡單，靈敏，快速的ROS檢測方法。 ROS檢測基於螢光探針DCFH-DA。 DCFH-DA是一種可滲透細胞的靈敏探針，用於檢測細胞內活性氧（reactive oxygen species , ROS）。 該DCFH-DA探針可在穿過活細胞膜時被細胞中的酯酶水解為DCFH。 DCFH沒有螢光並且不能透過細胞膜，然後可以被ROS氧化並產生螢光DCF。 然後可以通過流式細胞儀或螢光顯微鏡檢測螢光，並據此分析細胞中活性氧（CheKine™ Reactive Oxygen Species , ROS）的含量。

Oxidative stress plays an important role in the research of cardiovascular disease, diabetes, osteoporosis, stroke, inflammatory disease, and neurodegenerative diseases and cancer,etc. ROS detection will help determine how oxidative stress adjusts various intracellular pathways.

Usage notes	• Do not mix the components between different batch numbers and manufacturers; otherwise, the results may be abnormal. • Avoid bubbles while mixing or redissolving components. • Change pipette tips frequently to avoid cross contamination between components. • Ensure that all components and equipment are at the proper temperature before starting the experiment. • The working concentration of positive control H2O2 should be 100 μM (Recommend during 50-200 μM depend on the cell types ). The level of Reactive Oxygen Species (ROS) will increase significantly after stimulated 30 min-4 h. The level of Reactive Oxygen Species (ROS) maybe very different for different cells while stimulating with the positive control. The stimulating time and the concentration of positive control could be prolonged or increased, if there is no obvious rise after 30 min . Or shorten the time or reduce the concentration of positive control, while the fluorescent signal is too strong. • During the experiment, the fluorescent probe DCFH-DA can be diluted in 1: 2000-1: 5000, and make the final concentration of DCFH-DA to 2-5 μM if the signal of negative control also too strong. The time of probe incubating can be adjusted in 15-60 minutes, and minimize the exposure time while reading the film, and the exposure time of the control group and test groups should to be consistent. • Reactive Oxygen Species (ROS) positive control (H2O2) is only used as a sample as a positive control, and not all test groups need positive control. • Please wash away the unbound prode after incubating the prode , otherwise it will cause a high background. • Try to shorten the detection time after incubating the probe (except for stimulation time) to reduce various possible errors.

Description:
Internucleosomal DNA fragmentation is a hallmark of apoptosis in mammalian cells. BioVision’s Apo-BrdU-IHCTM Kit is a two-color TUNEL (Terminal deoxynucleotide transferase dUTP Nick End Labeling) assay for labeling DNA breaks to detect apoptotic cells by immunohistochemistry. The kit contains positive/negative control slides for assessing reagent performance; reaction and blocking buffers for processing individual steps in the assay; proteinase K; terminal deoxynucleotidyl tranferase enzyme (TdT), bromodeoxyuridine triphosphate (Br-dUTP), biotin labeled antiBrdU antibody for labeling DNA breaks, horseradish peroxidase streptavidin conjugate, DAB, H₂O₂/Urea tablets for color generation and methyl green solution for counter staining the cells.

PiColorLock is a phosphate detection reagent for measuring Phosphatases, ATPases and Nucleotidases.

 

Colorimetric assays for ATPases are invariably based on the formation of colored complexes between an inorganic phosphate and a dye molecule under acidic conditions. Such assays are beset with problems of reagent precipitation and high backgrounds caused by impure substrates (i.e. contaminated with inorganic phosphate) and/or by non-enzymatic (acid) hydrolysis of the substrate.

At the heart of our assay kits is PiColorLock Gold, a superior phosphate detection reagent which ensures high stability of the colored dye-phosphate complexes. Together with specially purified substrates and proprietary stabilizers our kits offer the lowest possible assay backgrounds and outstanding assay performance.

Uniquely, Innova Biosciences ATPase assay kits contain highly purified ATP, which have had any free inorganic phosphate removed – this means the assays have the lowest possible background signal.

PiColorLock reagent may also be purchased separately for developing simple assays for any phosphate-generating enzyme. It is offered in both high (Gold) and standard sensitivity (ALS) formulations to suit almost any assay situation.

Inorganic phosphate (Pi) is a product of many enzymatic reactions, for example, those catalysed by phosphatases and ATPases. Measurement of released Pi allows enzyme activity to be determined, but contamination of enzymes or buffers by Pi can lead to unacceptably high assay backgrounds.

PiBind™ resin provides a quick and easy way to remove contaminating Pi from buffers. The resin works over a broad range of pH values and is unaffected by many commonly used buffer additives. PiBind™ resin can also be used to remove Pi from protein samples (e.g. tissue extracts). This method is quicker than dialysis and unlike desalting does not necessarily lead to a significant dilution of the sample.

Description:Aspartate
aminotransferase (AST), also known as Glutamate-oxaloacetate transaminase (GOT)
is a transaminase (EC 2.6.1 .1) similar
to the more liver specific alanine transaminase (ALT). Although commonly
included clinically as part of a diagnostic liver function test, AST has a
broader clinical utility since it may also be elevated in diseases affecting
other organs, such as the heart or muscles in myocardial infarction, also in
acute pancreatitis, acute hemolytic anemia, severe burns, acute renal disease,
musculoskeletal diseases and trauma. It catalyzes the reaction:

Aspartate
+ ⍺-Ketoglutarate   ⇌ 
 Oxaloacetate + Glutamate  

 Diagnostically,
it is almost always measured in units/liter (U/L). In BioVision’s AST Assay
Kit, an amino group is transferred of from aspartate to ⍺-ketoglutarate.
The products of this reversible transamination reaction are oxaloacetate and
glutamate. The glutamate is detected in a reaction that concomitantly converts
a nearly colorless probe to color (λmax = 450 nm). The kit provides a rapid,
simple, sensitive and reliable test suitable as a high throughput activity
assay of AST with a detection limit of 10 mU per well.

Description:


Glutathione S-transferase (GST) is a family of enzymes that play an important
role in detoxification of xenobiotics. GST catalyzes the formation of the thiol
group of glutathione to electrophilic xenobiotics. It utilizes glutathione to
scavenge potentially toxic compounds including those produced as a result of
oxidative stress and is part of the defense mechanism against the mutagenic,
carcinogenic and toxic effects of such compounds. The GST Colorimetric Activity
Assay Kit is based upon the GST-catalyzed reaction between GSH and the GST
substrate, CDNB (1-chloro-2,4-dinitrobenzene, which has the broadest range of
isozyme detectability (e.g., alpha-, mu-, pi-, and other GST isoforms). Under
certain conditions, the interaction between glutathione and CDNB is totally
dependent on the presence of active GST. The GST-catalyzed formation of GS-DNB
produces a dinitrophenyl thioether which can be detected by spectrophotometer
at 340 nm. One unit of GST activity is defined as the amount of enzyme
producing 1 µmol of GS-DNB conjugate/min under the conditions of the assay. The
kit can detect GST activity in crude cell lysate or purified protein fraction,
and also quantitate GST-tagged fusion protein. Detect limit: Active GST < 4
mU.

Background Descriptions: Internucleosomal DNA fragmentation is a hallmark of apoptosis
in mammalian cells. BioVision’s Apo-BrdU In Situ DNA Fragmentation Assay Kit
provides complete components including positive and negative control cells for
conveniently detecting DNA fragmentation by fluorescence microscopy or flow
cytometry. The kit utilizes Br-dUTP (bromolated deoxyuridine triphosphate
nucleotides) which is more readily incorporated into DNA strand breaks than
other larger ligands (e.g., fluorescein, biotin or digoxigenin). The greater
incorporation gives rise to brighter signal when the Br-dUTP sites are
identified by a fluorescein labeled anti-BrdU monoclonal antibody.

conveniently detecting DNA fragmentation by fluorescence microscopy or flow
