內毒素檢測試劑(LAL Reagent) Gel-Colt(凝膠法) 鱟試劑與內毒素反應會產生凝膠，利用凝膠的生成與否來偵測內毒素的方法，為最早發展出來的LAL測試方法。

簡單省時的過程





每一個測試應該包括一個陽性對照和一個陰性對照 (LAL試劑)。應用實例如下：





1. 樣品：1 mg/ml的蛋白質A，被置於pH 7.4的磷酸緩衝液中。蛋白質A是由Ni-NTA樹脂純化大腸桿菌得到。





2. 按照稀釋比例1: 200,000，1: 400,000，1: 800,000在LAL試劑水中稀釋。





3.該測試是在上述條件下進行，得到的結果如下：





4.初步判斷該樣品的內毒素在50,000到100,000 EU/ml之間

Endotoxin Assay Kits

Technical Advantages:

Under the standard conditions, the absorbance at 545 nm shows a linear relationship with the concentration in range of both 0.01 - 0.1 EU/ml and 0.1 - 1 EU/ml. Plot the absorbance for the four standards on the x-axis, the corresponding endotoxin concentration in EU/ml on the y-axis. Draw a best-fit straight line among these points and determine endotoxin concentrations of samples graphically.

If the mean absorbance of a sample is x, the endotoxin concentration of sample will be 0.2618x – 0.0012 EU/ml. All incubations were performed for 45 min. Above is only an example curve show, and the OD values of standards may be different with different assays.
