RHPS4 是一種端粒酶抑制劑(telomerase inhibitor)。染色體末端(chromosome termini)的保護、降解(degradation)和重組(recombination)受端粒(telomeres)調控。最近的一個模型提出端粒(telomeres)可以在染色體末端(chromosome end)形成一個cap。因此，端粒帽的完整性(telomere cap integrity)應該是完整的，以允許細胞分裂(cell division)繼續進行。體外試驗：發現 RHPS4 對 UXF1138L 細胞的處理導致端粒酶催化亞基 (telomerase catalytic subunit, hTERT) 從細胞核中置換出來，誘導端粒啟動的 DNA 損傷信號傳導(f telomere-initiated DNA-damage signaling)以及染色體融合(chromosome fusions)。進一步報導了 RHPS4 對以集落形式(colonies)生長的癌細胞比以單層形式(monolayers)生長的細胞更有效。此外，人臍帶血(human cord blood)和HEK293T胚胎腎細胞(HEK293T embryonic kidney cell)集落的形成單位對RHPS4的抵抗力更強[1]。體內試驗：動物研究發現，與對照組相比，經 RHPS4 處理的 UXF1138L 異種移植物的克隆形成性降低、核 hTERT 表達缺失和有絲分裂異常的誘導。儘管單獨的 RHPS4 顯示出有限的體內功效，但與有絲分裂紡錘體毒物紫杉醇的聯合治療可導致腫瘤緩解並進一步增強端粒功能障礙 [1]。
