透析膜需選擇之項目 (透析膜材質、分子量、FW平寬)

  1. 透析膜材質 / CE膜或RC膜選擇| 請先參閱 Spectrum透析膜與化學溶液相溶性比較表
  2. 選擇透析膜MWCO大小＝欲保留分子量大小÷3。例如欲保留50KD以上物質，可選擇MWCO 20KD的透析膜。
  3. FW平寬。透析膜平寬（FW）若為16 mm，體積／長度比為79 ml/cm。

原理介紹透析之原理介紹 | 如何選擇適合的分子量

利用分子會從高濃度擴散到低濃度，將待透析液放在具分子大小選擇性的透析膜中，進行物質交換。此時大分子的溶質會被保留，而小分子溶質會通過透析膜移除，最終達到去除的效果。一般而言，常規的透析需要進行2天的時間，才能達到透析的目的。而擴散係數(diffusion coefficients)、pH、溫度、時間、物質濃度、溶液體積、緩衝液體積，緩衝液更換次數、透析膜表面積、透析膜厚度，攪拌等因素都會影響到透析的效率。

MWCO是甚麼？

Molecular Weight Cut Off（MWCO）載留分子量是評估透析膜孔洞大小的單位。MWCO被定義為經過17小時透析後，保留90%以上的分子量大小。
選擇透析膜MWCO大小＝欲保留分子量大小÷3。例如欲保留50KD以上物質，可選擇MWCO 20KD的透析膜。

使用方法A如何進行透析：

將待透析品放入透析膜後，置入100X體積之緩衝液後開始進行透析。透析中至少進行三次緩衝液的換置，操作時間至少為16－24小時：

    • 第一次：透析進行2－3小時後。
    • 第一次：透析進行4－5小時後。
    • 最後一次：overnight之前。

使用方法B透析膜需要前處理嗎?

  1. Biotech RC、CE、與PVDF透析膜使用DI水清洗15－30分鐘（移除保存液sodium azide殘留）。
  2. Spectra/Por® 7 Standard RC透析膜已經過重金屬與硫化物去除的處理，只需要浸泡在DI水中15－30分鐘即可（移除保存液sodium azide殘留）。
  3. Spectra/Por 1－6 Standard RC透析膜，針對不同需求會有額外的處理手續。一般而言，可使用水清潔移除Spectra/Por 1－6 Standard RC透析膜中甘油的殘留。

Standard Grade Regenerated Cellulose (RC) membrane 為透明、富有彈性的透析袋；擁有極高的化學耐受性，可承受低濃度的強酸強鹼溶液、高濃度的弱酸弱鹼溶液、多數醇類和稀釋的有機溶劑 (ex:DMSO)，並可在承受 4-121 °C 和 pH 2-12 的環境下進行實驗；適用於將分子量差異大的樣品進行分離實驗，包含樣品製備、去除鹽類、置換緩衝溶液、調整pH值與蛋白質純化等。Spectra/Por ® 1 - 5 Standard RC Dialysis Tubing 為方便使用者保存，以乾燥的型式包裝出貨。一卷為 15 或 30 公尺，依需求自行裁切成合適的長度，對實驗需求量大者，是較為經濟型的包裝。大部分乾燥的透析袋在實驗前需浸泡過濾水30分鐘以去除保濕劑-glycerin。若乾燥的透析膜不帶有glycerin則只要稍做潤濕即可使用

原理介紹透析膜長度

全長＝［（樣品容量）／（體積／長度比）］＋（額外的10－20%）＋4 cm →計算您要使用多長的透析膜例如：透析膜平寬（FW）為16 mm，體積／長度比為0.79 ml/cm。欲透析5 ml樣品，則至少需約6.5 cm透析膜。但還需要額外的10－20%的長度（至少1 cm）使透析管填充空氣維持浮力。最後還需要頭尾各2 cm給透析夾使用。整個透析膜長至少需要11.5 cm長。

原理介紹透析夾之選擇

Universal透析夾適用各種透析膜，Speatra/Por僅適用於RC透析膜 →透析夾詳細資訊 使用時選用大於透析膜平寬（Flat Width, FW）4－10mm以上的透析夾來進行密封。

Spectra/Por Closures  RC材質透析膜專用夾 

此專用夾的夾力較大與緊密，因此適合RC材質的透析膜(軟與薄)，並有輕巧的標準型夾一般可懸浮於透析液表面; 加重型可沉於透析液底部，與前者搭配可使透析膜垂直於透析液中，及磁力加重型透析夾含磁鐵可同時當磁石攪拌使用。

標準夾 (Standard Closure Type)

由於標準夾可漂浮，使用時夾於透析膜管上方增加浮力。可耐高溫高壓滅菌，並擁有多種顏色做區分。

重量夾 (Weighted Closure Type)

可滅菌之重量夾含有以fluorocarbon 包覆表面的不鏽鋼棒。固定於透析膜管下方使固定透析膜管的垂直方向。

磁力加重夾 (Magnetic Closure Type)

可滅菌之磁力加重夾含有以fluorocarbon 包覆表面的不鏽鋼棒，固定於透析膜管下方，可當作傳統的攪拌磁石
(stir bars)使用，因此在透析過程中無須而外再放入攪拌磁石。此方式透析膜會完全沉入透析液中，並將容器
置於磁力攪拌器 (stir plate) 上，利用溫和的轉速進行透析實驗。

標準夾和重量夾配對組 (Paired Standard & Weighted Closures)

利用標準夾和重量夾一上一下使用，使透析膜管達到浮起並垂直於透析液中，兩種透析夾均可進行滅菌處理。

AbSelect Antibody Purification System (860-0010)

Innova Bioseiences提供一系統抗體純化套組，每組套組包括抗體純化之buffer及resin，有效純化不同來源之抗體含量。其純化來源，可為市售抗體之純化(貨號#860-0010/rabbit; 貨號#830-0010/mouse; 貨號#890-0010/rat/sheep/human/goat)、腹水及血清產製抗體之純化(貨號#863-0500/rabbit; 貨號#893-0500/rat/sheep/human/goat/mouse)、融合瘤組織培養液(TCS)產製之抗體(貨號#862-0500/rabbit/human; 貨號#832-0500/mouse)、低濃度抗體之純化及濃縮(貨號#861-0010)。有效移除溶液中BSA、Glycine、Tris、Azide等殘留分子，純化後抗體溶液，可直接應用於lateral flow test奈米金抗體標誌套組 (Innova gold,#230-0010, innova biosciences)、與HRP/螢光抗體標誌套組(Innova lightning-link, #701-0000)。

抗體濃縮及Clean up系統

適合應用: 低濃度抗體之純化及濃縮。

干擾酵素型HRP、Biotin、streptavidin及FITC螢光等之抗體標誌反應。移除溶液中殘留之amino group。純化後之抗體，可直接進行奈米金抗體標誌套組 (Innovagold,#230-0010, innova biosciences)及抗體螢光或HRP標誌(lightning_link)。

抗體濃縮及Clean up流程

Step 1 加入 0.5ml 以上的抗體量至 spin cartridge.

Step 2 15,000g離心 1 to 3 分, 使抗體量降低至100ul

Step 3 吸掉離心管的溶液

Step 4 加入 400μl conjugation buffer至spin
cartridge中的抗體

Step 5 15,000g離心 1 to 3 分, 使抗體量降低至100ul

Step 6吸掉離心管的溶液.

Step 7 重覆1-6步驟, 置換抗體溶液。

Step 8 由spin cartridge中回收純心後的抗體

 

Antibody Concentration and Cleanup Kit

Antibodies are sometimes only available at low concentrations and often contain low molecular weight substances that interfere in labeling reactions with enzymes, biotin, streptavidin and fluorophores. The Antibody Concentration and Clean Up Kit allows for the quick and easy concentration of antibodies and proteins. The kit can also be used to reduce the concentration of many unwanted additives often found in antibody formulations such as azide, glycine or tris.

The antibody clean up kit method utilizes a simple spin
column to easily and quickly remove excess buffer from the antibody thereby providing a more concentrated antibody solution. The clean up kit also allows the experimenter to perform a simple buffer exchange to transfer the antibody into a more favorable conjugation buffer.

 

DIY抗體標誌螢光(FITC,PE,Cy3,Cy5,Cy7 dye)

注意事項

請確認抗體的保存條件。傳統市售的抗體標誌套組，在使用前皆需dialysis純化，將抗體保存的物質移除。Lightning-link抗體標誌套組在大部份的保存條件下，不需要再dialysis的步驟，例如一般salts (e.g. NaCl)、sugars(e.g. sucrose)、及chelators (e.g. EDTA)不會影響標誌的活性。大部的市售的抗體保存在1%BSA、0.5%Azide及0.5% Gelatin 輕微或幾乎不會影響標誌的活性。在少數情況下，若是您購買的抗體保存在amino acids(e.g. glycine)、blockers(e.g. ethanolamine)及thiols(DTT, mercaptoethanol)情況下，記得純化您的抗體，(abselect抗體純化套組) 避免抑制您的標誌活性了!

Chickens were immunized with the E-Tag peptide
(GAPVPYPDPLEPR) coupled to KLH. After multiple immunizations in Freund's
adjuvant, eggs were collected and the immunoglobulin fraction prepared.
Antibodies were immunoaffinity purified using the peptide immobilized on a
solid support and then conjugated to biotin.

This antibody is suitable for ELISA and Western
blots. Optimal working dilutions should be determined for your particular
assay conditions.

Description




Immunoglobulin G (Fc) Specific is a IgG is a monomeric immunoglobulin,
built of two heavy chains gamma and two light chains. Each molecule has two
antigen binding sites. This is the most abundant immunoglobulin and is
approximately equally distributed in blood and in tissue liquids,
constituting 75% of serum immunoglobulins in humans. This is the only isotype
that can pass through the human placenta, thereby providing protection to the
fetus in its first weeks of life before its own immune system has developed.
It can bind to many kinds of pathogens, for example viruses, bacteria, and
fungi, and protects the body against them by complement activation (classic
pathway), opsonization for phagocytosis and neutralisation of their toxins.
There are 4 subclasses: IgG1 (66%), IgG2 (23%), IgG3 (7%) and IgG4 (4%).

Description




M13, fd, F1 Filamentous Phages -HRP is a The bacteriophage known as
"M13" forms the basis of cloning systems designed to easily
introduce mutations into genes inserted into the phage genome. It also has
been used in various "phage display" methodologies and combinatorial
DNA and peptide libraries. Coat protein A (G3P) is necessary for adsorption
of the virion onto the F-pilus of the host cell. There are about five copies
of this protein per mature phage. Consists of three domains (N1, N2, and CT).
The N2 domain interacts with the F pilus, whereas the N1 domain (connected to
N2 by a flexible glycine-rich linker and tightly interacting with it on the
phage) forms a complex with the C-terminal domain of tolA at later stages of
the infection process. They are located at the adsorption end of the phage

內毒素來源為革蘭式陰性菌細胞壁。成分為脂多醣體 (Lipopolysaccharide, LPS)。其結構組成：1. 脂質A (lipid A) 2. 多醣類核心 (core polysaccharides) 3. O-多醣類 (O-polysaccharide)





引發症狀：血管內凝集血栓症、敗血性休克





Endotoxin (LPS) is released
primarily upon cell lysis when bacterial cell dies can make up the majority of
pyrogens. Endotoxin removal is especially important when performing
immunological readouts and developing manufacturing processes because endotoxin
can cause false readings in cell based assays and there are limits to the
amount of endotoxin allowed in human products. It is always desirable to
rapidly and effectively remove pyrogen from pharmaceutical products,
recombinant proteins produced in E. coli (or other gram negative
bacteria), and tissue culture media.
