Nivolumab，也稱為 Opdivo，是一種單克隆抗體(monoclonal antibody)，用於通過抑制稱為程序性死亡-1 (PD-1) 的細胞途徑來治療晚期非小細胞肺癌 (non-small cell lung cancer , NSCLC)。Nivolumab與T細胞表面的PD-1結合，從而阻斷PD-1/PD-2觸發的免疫抑制信號通路，恢復T細胞(antitumor function)的抗腫瘤功能。Nivolumab以高特異性結合 PD-1 [1]，納武單抗可有效抑制 PD-1 與其配體（PD-L1 和 PD-L2，IC50 值分別為 2.52 和 2.59 nmol/L）的相互作用。在 CHO 細胞中，使用 FACS 評估配體與 PD-1 的結合，Nivolumab單抗介導的 PD-1 與 PD-L1 結合的抑制（IC50 值為 1.04 nmol/L）和 PD-L2（IC50 值）之間沒有明顯差異0.97 nmol/L)。此外，當缺乏抗原或 T 細胞受體刺激時，nivolumab沒有刺激作用，不會引起非特異性淋巴細胞活化(nonspecific lymphocyte activation) [2]。在cynomolgus monkey模型中使用 10 和 50 mg/kg Nivolumab (i.v.)，血清 nivolumab 似乎沒有明顯變化，也沒有明顯的性別差異。用 50 mg/kg nivolumab治療後，T4 和 TSH 水平沒有變化，但 T3 在女性中可逆地下降了 28%，而在男性中，T3、T4 或 TSH 水平沒有變化 [2]。nivolumab在猴子身上可能沒有毒性，但在人體臨床試驗中已經觀察到這種毒性，nivolumab在 I 期試驗中具有良好的安全性 [1]。一個值得研究的問題是，在 PD-1 缺陷小鼠中觀察到的肺炎是 MRL 遺傳背景而不是其他遺傳背景引起的 [2]。
