【Ni-IDA高親和力純化管柱】

Ni-IDA高親和力純化產品(G239)是一種用於純化任何重組表達系統，如細菌、酵母、昆蟲和哺乳動物細胞等表達系統表達的6xHis標籤的重組蛋白的瓊脂糖純化介質，它是通過三個結合位點與一種三齒螯合劑（亞氨基二乙酸）共價偶聯，該螯合劑可螯合Ni2+。該純化介質可在天然、變性或雜交條件下純化his標誌重組蛋白。結合到純化介質上的蛋白，用低pH緩衝液或咪唑溶液、甚至組氨酸溶液都可洗脫下來。

GST標誌重組蛋白純化

 

 

 

 

 

 

 

 

 

 

親和層析法為廣泛應用的蛋白質純化方法之一。使用His-Tag 或是GST-Tag 純化的流程相當簡單，只要經過Loading、Washing、Elute這三個步驟就可以得到一定純度以上的蛋白質。搭配FPLC 儀器進行純化，可獲得更高純度或更大量的重組蛋白。

 

 

 

 

 

【GST-Tag 純化原理】GST-Tag fusion protein 是使用glutathione S-transferase (GST)與glutathione 的親和特性（酵素和基質）進行重組蛋白質純化。Glutathione 在進行蛋白質純化前預先 coating 在Agarose beads 上作為glutathione-Agarose resin，當GST-fusion protein 通過glutathione-Agarose resin 時，GST-Tag 與glutathione 會產生親和性反應，並結合在glutathione coated resin 上，此時未含GST 的蛋白質會通過resin 並使用wash buffer 達到純化目的。最後在elute 步驟中放入含有free glutathione 的elute buffer，就能使GST-protein 脫離resin 並溶解於elute buffer。

Description

Affinity chromatography is a method of separating biochemical mixtures based on highly specific molecular interactions. The Ni-IDA agarose is developed for the purification of proteins with an affinity tag of six consecutive histidine residues. This histidine-metal ion interaction makes one-step purification possible for proteins from any expression system, under native or denaturing conditions. The His-tag sequence binds to Ni²⁺ cations, which are immobilized onto a solid support using iminodiacetic acid groups (IDA). Impurities are removed and the purified His-tagged proteins can be eluted by imidazole or a reduced pH solution.

產品介紹

lysozyme 可水解細菌細胞壁中 peptidoglycan 的糖鍵，使得菌體無法承受滲透壓逆境而破裂，可改良蛋白質過程造成的damage；但加入lysozyme前要先加入cell extraction buffer。最佳化狀態須加入protease inhibitors,及lysozyme 可以降低蛋白質降解粘稠度及改良萃取過程中的傷害。 EZLys(TM)菌液萃取蛋白質試劑, 500ml，其可以萃取native及non-denatured form蛋白質。產品有提供soluble片段及inclusion body片段的操作方法。如附件所示。

 

A proprietary formulation prepared in a 50 mM Tris pH8.0 buffer and designed for gentle

disruption of cell membrane to extract proteins. It provides a simple, rapid, low cost method for releasing expressed target protein for preparation, purification, or other applications. The EZLys™ reagent is an optically transparent, non-denaturing formulation which can be easily removed by dialysis. It is also useful for the preparation of high purity inclusion bodies for subsequent purification. This reagent does not interfere with typical protein immobilization or cross linking and is suitable for use in the BCA or Bradford protein assays. Ideal pH range is pH 5 -10. The 100 ml and 500 ml reagent are sufficient for protein extraction from 25 g and 125 g of cell paste.
