One
of the most reliable, quantitative assays for the suppression of target genes
by a specific miRNA is the utilization of a reporter gene like luciferase or
GFP. Target site for a specific miRNA is subcloned downstream of the reporter
gene and co-transfected along with a miRNA expressing vector into cells.
Inhibition of reporter genes by the expressed miRNA as compared with valid
controls proves the regulation of genes with target site at 3'-UTR by the
specific miRNA. To facilitate your research on genes of your interests, we have
developed 3'-UTR library of lentiviral vectors for all human genes down-stream
a reporter gene of either luciferase or GFP. The advantage of lentiviral vector
is its flexibility for either transfection or viral vector infection
